A Novel Regulation Of Adrenomedullin Receptor By PDGF In Mesangial Cells- Role Of Receptor Activity Modifying Protein-3
نویسندگان
چکیده
Receptor activity modifying protein-3 (RAMP-3) has been shown to complex with the calcitonin receptor-like receptor establishing a functional receptor for adrenomedullin (AM). AM exhibits potent anti-proliferative and anti-migratory effects on rat mesangial cells (RMC). In this study we investigated the effect of platelet-derived growth factor (PDGF) on RAMP-3 expression in RMC. We show here that PDGF-BB stimulates RAMP-3 mRNA expression in a concentration-dependent manner. Pre-treatment with actinomycin-D and α-amanitin demonstrates that this effect is independent of new RNA synthesis. Furthermore, PDGF increased the half-life of RAMP-3 mRNA from 66.5 to 331.6 min. Using selective inhibitors, our results also indicate that the increase in RAMP3 mRNA is MAPKK/MAPK and p38 MAPK dependent. PDGF also caused a corresponding elevation in membrane-associated RAMP-3 protein. Associated with this increase, PDGF pretreatment led to a significantly higher AM-mediated adenylate cyclase activity, suggesting a functional consequence for the PDGF-induced increase in RAMP-3 expression. Taken together, these data identify PDGF-dependent regulation of RAMP-3 expression as a possible mechanism for modulating mesangial cell’s responsiveness to AM.

متن کاملThe Effect of Aspartate-Lysine-Isoleucine and Aspartate-Arginine-Tyrosine Mutations on the Expression and Activity of Vasopressin V2 Receptor Gene
Background: Vasopressin type 2 receptor (V2R) plays an important role in the water reabsorption in the kidney collecting ducts. V2R is a G protein coupled receptor (GPCR) and the triplet of amino acids aspartate-arginine-histidine (DRH) in this receptor might significantly influence its activity similar to other GPCR. However, the role of this motif has not been fully confirmed. Therefore, the ...
متن کاملPI-3-kinase and MAPK regulate mesangial cell proliferation and migration in response to PDGF.
Proliferation and migration are important biological responses of mesangial cells to injury. Platelet-derived growth factor (PDGF) is a prime candidate to mediate these responses in glomerular disease. PDGF and its receptor (PDGFR) are upregulated in the mesangium during glomerular injury. We have recently shown that PDGF activates phosphatidylinositol 3-kinase (PI-3-kinase) in cultured mesangi...

We investigated whether platelet-derived growth factor (PDGF), or its receptor (PDGF-R), was upregulated in a rat model of mesangial proliferative glomerulonephritis. A marked increase in both PDGF A- and B-chain mRNA could be demonstrated in glomerular RNA by Northern blot analysis 3 and 5 days after disease induction, corresponding to the time of mesangial cell proliferation. PDGF-R beta-subu...
متن کاملA novel treatment approach for retinoblastoma by targeting epithelial growth factor receptor expression with a shRNA lentiviral system
Objective(s): Non-invasive treatment options for retinoblastoma (RB), the most common malignant eye tumor among children, are lacking. Epithelial growth factor receptor (EGFR) accelerates cell proliferation, survival, and invasion of many tumors including RB. However, RB treatment by targeting EGFR has not yet been researched. In the current study, we investigated the effect of EGFR down-regula...
